I have used the 5D now for 2 years, taken a good number of shots and generally given it hell. It has withstood everything I threw at it, sand on the Gallapagos islands, salt and bird doodoo on the Farnes, rain and sleet in the UK and freezing temperatures here in Germany. It still goes strong, image quality and detail is amazing due to the weak AA filter.
I really do not want to get rid of it.

After using Canon SLR for many years I waited for the right DSLR to come along. I wanted something that felt solid like my EOS 50E did, yet had full frame so my wide angle lenses stayed wide.
So when the 5D came out I was quite excited. I did consider the 20/30D at the time as well. However the 5D won purely on the full frame (and thus larger viewfinder).
Build is superb, O.k. its not 1D quality, but its also not 1D price either. 3 frames per second isnt great either, but for me it does what I want. Lack of flash didnt bother me as I already have an external flash. To be honest, Ive never really rated built in flash other than for a fill-in flash.
Currently the 5D can be bought as a kit with the 24-105. This is superb value as this lens works well with the 5D.
Dust can be an issue with the 5D, however it seems dust prevention methods arent always that great at removing dust. For me dust isnt a big problem as I tend to clone it out in post processing (using Adobe Lightroom).
Age may be a factor for some with the 5D. It will be 2 years old in September, however it will not become useless overnight. I plan to keep hold of my 5D for a good while yet.

I bought the 5D to shoot weddings with, and it is fantastic for it. The picture quality is superb, and the high ISO performance very good. The camera is relatively light, compared to its 1D big brothers, which is another plus when youre using it all day. And of course it is full frame, so you get a decent wide angle without needing a ultra wide angle lens like you would on a camera with a smaller sensor.
I would thoroughly recommend this camera for people shooting weddings, portraits or landscapes. However, if your main interest is in nature or sports photography, you will probably find its 3 frames per second a little too slow for you.
Of course, it isnt perfect. There are three things I would look for in a successor to the 5D to improve it:
1) Increased dynamic range (not a weakness specific to the 5D, but a general area for improvement in DSLRs).
2) Integrated sensor cleaning (dist is an issue if you change lenses regularly).
3) ISO setting indicator in the viewfinder, alongside the shutter speed and aperture.
All in all, I think the 5D is great, and Ive not regetted buying it.
